would like to know if I just plug the return line at the fuel tank and the three vapor lines in the trunk ;for Iam using 32/36DFEV carb on my 78 spider.This should not cause any problems in the trunk area

You want to close the three vapor lines up? Those lines vent the tank. You'll end up starving for fuel and collapsing the tank if you plug them.

What if I don`t plug up the lines ,run off the single line in the engine bay a fuel filter and plug into the intake mainfold.Has anyone done this.

Fuel pumps, whether electric or mechanical, work better and last longer with a return line. You can run one line up and one line back, plugging the rest, but as Denise says, you'll need to vent the tank somehow. One popular way is to drill a tiny hole in the gas cap.
Of course, this setup will flunk any emissions inspection, perhaps degrading the resale value of your car.
There are two evaporative emissions systems on your '78. You can leave the fuel tank system intact. On the exploded diagram I have of the 32/36DFEV, there seems to be a provision for a return line, so nothing changes from the OE carb except the evap line from the engine compartment canister has nowhere to go on the DFEV. I would think that you could simply plug this line, and you'd be good to go. I routed this line to a non-functioning nipple on my 38DGS, and it looks "correct" to a would-be inspector.
